When the effect of calcium on the oxidation of linoleic acid by potato tuber 5-lipoxygenase (LOX) was investigated, it was seen to promote the enzyme's activity at pH values higher than the optimum pH of 6.3, resulting in an enzyme activation at alkaline pH. Kinetic analysis of calcium activation at different pH values revealed that the cation abolished the inhibition by high substrate concentration, which occurs in the absence of Ca(2+), thus leading to activation at high substrate concentration. Studies were conducted to investigate the influence of Ca(2+) on the physicochemical nature of the substrate and its effect on the LOX activity expression. It was concluded that the aggregation mode rather than the aggregation state of linoleic acid is responsible for potato 5-LOX changes.  相似文献

The Sinorhizobium -legume interaction is sensitive to a number of environmental factors, soil acidity being one of the most important. In the typic Hapludoll soil of the central-southern region of Córdoba (Argentine) it was found that the nodulation of alfalfa ( Medicago sativaL.) roots was impaired with a reduction in shoot dry matter under conditions of soil acidity (pH 5.5) Our results showed that the addition of lime as dolomitic limestone at a concentration of 1 t ha -1to acid soil caused a significant increase in the nodulation of alfalfa roots inoculated with the strain Sinorhizobium meliloti SEMIA 116 (recommended inoculum for alfalfa) in the greenhouse experiments. The success of the lime treatment may be related not only to an increase in the pH values but also to an increase in the Ca concentration, improving the growth of S. meliloti and its nodulation ability under acidic conditions. In this study, we also demonstrated an increment in the bacterial growth rate as well as in the production of exopolysaccharides and lipopolysaccharides under low pH (5.5) and a high Ca concentration (5 mM) in the culture medium.  相似文献

Laboratory studies were undertaken to evaluate the influence of fertilizers on pendimethalin volatilization and persistence in soil. Various organic fertilizers such as liquid humic substances and urea were used at doses of 100 L/ha or 170 kg of N/ha, respectively. Herbicide residues were determined in air, soil solution, and soil samples by GC-ECD; the recovery of pendimethalin from spiked fertilized or control samples was found to be 81-103%. Liquid humic fertilizers increased pendimethalin dissipation during the first part of the assay, although 4 months after application, herbicide levels were similar to those observed in unfertilized soil. Fertilization of soil with urea decreased, in general, pendimethalin volatilization but increased herbicide levels in soil solution and persistence in soil, with a pendimethalin half-life approximately 70% higher than that found in unfertilized soil.  相似文献

Bitter taste, an organoleptic characteristic of virgin olive oil, has been related to phenolic compound composition. The usual method to assess this attribute is by a sensorial panel of tasters, while in the laboratory; methods based on physicochemical properties have been assayed as K225, the most widely used one. However, a direct determination of bitterness in virgin olive oil is useful for quality-control purposes. The proposed method is supported by the observable spectral change undergone by the compounds responsible for bitterness as pH varied. This measurement was carried out directly in the oil, without prior isolation of bitter analytes. The difference of absorbance between alkaline and neutral medium showed a highly significant correlation (r = 0.988, p < 0.0001) with the conventional parameter (K225). The method was rapid, required a small sample, allowed direct determination of bitterness in virgin olive oil, and could be easily automated.  相似文献

The residual effects of adding 40 t ha–1 sewage sludge (SL) to a degraded soil cropped with barley were investigated after 9 and 36 months in a field experiment under semiarid conditions. The principal soil properties were apparently still affected by SL amendment 9 months after application but the effects disappeared after 36 months. With respect to control soil humic acids (HAs), the SL-HA was characterized by higher contents of S- and N-containing groups, smaller contents of acidic groups, a prevalent aliphaticity, extended molecular heterogeneity, and smaller degrees of aromatic polycondensation and humification. Amendment with SL caused an increase in N, H, S and aliphaticity contents and a decrease in C/N ratios and O and acidic functional group contents in soil HAs isolated 9 months after SL application. These effects tended to decrease after 36 months, most probably because the slightly humified SL-HA was mineralised over time through extended microbial oxidation, while only the most recalcitrant components such as S-containing and aromatic structures were partially accumulated by incorporation into soil HA. Microbial biomass, basal respiration, metabolic quotient and enzymatic activities increased in soil 9 months after SL application, possibly because of increased soil microbial metabolism and enhanced mineralisation processes. After 36 months these properties returned to values similar to those of the unamended soil, presumably due to the loss of energy sources.  相似文献

Studies of Cd and Zn sorption using Na-saturated kaolinite and montmorillonite, and low metal solution concentrations similar to those found in the environment, showed that metal sorption affinity (measured by K d values) decreased markedly with increasing surface metal loading for both layer silicates. For equilibrium solution concentrations <0.1 μmol L?1 for Cd, and < 1 μmol L?1 for Zn, both metals were sorbed with greater affinity by kaolinite than montmorillonite. These results were probably due to the higher proportion of weakly acidic edge sites present on kaolinite surfaces. In the case of Zn there was an affinity reversal for equilibrium solution concentrations > 1 μmol L?1, which was attributed to the permanent charge sites of montmorillonite. Cadmium ions were sorbed, by kaolinite, with greater affinity than Zn for equilibrium solution concentrations between 0.3 to 1.5 μmol L?1. This result was attributed to retention of these metal ions through electrostatic attraction by permanent charge sites present on the kaolinite used in this work. According of these results it seems that metal sorption by these layer silicates involves predominantly edge weak acid sites at lower surface coverages (higher affinity sites), and permanent charge sites at higher metal coverages (lower affinity sites). It was concluded that Cd and Zn sorption by those two layer silicates is greatly influenced by surface metal coverage, and results cannot be extrapolated from low to high surface coverages, and viceversa.  相似文献

A lipoxygenase from Terfezia claveryi Chatin ascocarp, a mycorrhizal hypogeous fungus, is described for the first time. The higher proportion of PUFA in T. claveryi ascocarps makes lipid rancidity the main factor limiting its storage life. Thus, the studies on LOX from T. claveryi are important because this enzyme, among other roles, may be involved in an alteration of lipids leading to consumer rejection. The enzyme has been purified to apparent homogeneity by phase partitioning in the presence of Triton X-114, followed by two steps of cation-exchange chromatography. The purified T. claveryi LOX preparation consisted of a single major band with an apparent molecular mass of 66 kDa after sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The enzymic activity exhibited a strong specificity toward linoleic and linolenic acids as substrates, while only 32% activity was observed using gamma-linolenic acid. The pH optimum of this enzyme was pH 7.0. When the enzyme reacted with linoleic acid, it produced a single peak, which comigrated with standard 13-hydroperoxy-octadecadienoic acid; 13-hydroperoxy-octadecatrienoic acid was produced during the reaction with linolenic acid.  相似文献

In this work, a single flow injection multicommuted system using solid-surface fluorescence spectroscopy has been explored for the determination of diphenylamine in apples and pears. The native fluorescence signal of diphenylamine retained on the solid support (C18 silica gel) was used for its determination (lambda(exc)/lambda(em) 291/372 nm). The sample treatment consists of a liquid-liquid extraction with acetonitrile followed by a cleanup step using primary-secondary amines. The use of a continuous flow assembly implementing multicommutation, based on a set of three-way solenoid valves controlled by appropriate software, provides the automatic control of sample, carrier, and eluting solution with remarkable advantages in relation to conventional flow injection approaches. Using an optimized sampling time, the proposed method was linear in the range 0.25-5 mg kg(-1) with a detection limit of 0.06 mg kg(-1) and RSD (percent) values better than 3%. The proposed method was successfully applied to the analysis of diphenylamine in different apple and pear samples fortified at different concentrations, and recoveries between 78 and 104% were found. The results obtained illustrate the usefulness of the proposed method for the screening and evaluation of postharvest treatment of crops possibly containing diphenylamine.  相似文献

Changes in phenolic compounds, total glucosinolates, and vitamin C were monitored during the productive period along five inflorescence development stages of three broccoli commercial cultivars (Marathon, Monterrey, and Vencedor). In an attempt to identify differences due to agronomic factors, broccoli cultivars were grown under different sulfur fertilization with poor (15 kg/ha) and rich (150 kg/ha) rates. Phenolic compounds and vitamin C concentrations showed, in all broccoli cultivars, a rising trend from the first stage until the over-maturity stage, both for rich and poor sulfur fertilization. Significant differences were detected in the first two stages between rich and poor sulfur fertilization in total glucosinolates for all broccoli cultivars, where the highest concentration was always observed in the second development stage (used as minimally processed product) during poor fertilization. With regard to the last three stages, the glucosinolate concentration in the poor sulfur fertilization started to slope down until the over-maturity stage. Where rich sulfur fertilization is concerned, the highest level was reached during the third stage (used as minimally processed product also), and after that, glucosinolate concentration decreased until the fifth stage.  相似文献

Dietary fiber (DF) intakes in Western countries only accounts for about one-third of the substrates required for colonic bacterial cell turnover. There is a general trend among nutritionists to extend the DF concept to include all food constituents reaching the colon. In this line, a method to quantify the major nondigestible components in plant foods, namely, the indigestible fraction (IF), is presented. Analytical conditions for IF determination are close to physiological. Samples, analyzed as eaten, were successively incubated with pepsin and alpha-amylase; after centrifugation and dialysis, insoluble and soluble IFs were obtained. IF values include DF, resistant starch, resistant protein, and other associated compounds. IF contents determined in common foods (cereals, legumes, vegetables, and fruits) were higher than DF contents. Calculated IF intakes were close to the estimated amount of substrates reaching the colon. IF data could be more useful than DF data from a nutritional point of view; therefore, IF is proposed as an alternative to DF for food labeling and food composition tables.  相似文献
